Studies on the Spontaneous Oxidation of Cysteine I. the Preparation of Iron-free Cystine and Cysteine Hydrochlorides*

چکیده
In carrying out studies on the oxidation of cysteine, it is most important that cysteine be used which is known to be completely free from contaminating metals such as iron and copper, both of which are powerful catalysts to this type of oxidation. In a quantitative study of this action, Mathews and Walker (1) showed that this reaction was sensitive to iron in amounts as small as 0.00001 M concentration. This catalytic activity of traces of iron on cysteine oxidation is employed by Warburg (2) in a method devised to measure amounts of iron too small to be detected by the usual methods. Because of the ubiquitous nature of iron, and also of copper, the preparation of a metal-free cysteine has been extremely difficult. The starting point is usually Lcystine, which is quite easily prepared from the keratins such as horn, feathers, hair, or wool by acid hydrolysis, with subsequent separation from the decolorized hydrolysate by careful neutralization. The cystine, which is obtained as typical hexagonal plates on recrystallization, is then reduced, usually by the tin and hydrochloric acid method described by Baumann (3) and by Friedmann (4). Cysteine Oxidation Reactions Catalyzed by a Mononuclear Non-heme Iron Enzyme (OvoA) in Ovothiol Biosynthesis
OvoA in ovothiol biosynthesis is a mononuclear non-heme iron enzyme catalyzing the oxidative coupling between histidine and cysteine. It can also catalyze the oxidative coupling between hercynine and cysteine, yet with a different regio-selectivity. Due to the potential application of this reaction for industrial ergothioneine production, in this study, we systematically characterized OvoA by a...
متن کاملDevelopment of a Method for measuring Reactive Oxygen Radicals Levels In Vitro and Study the Effects of Vitamin C and E on Radical Production Reaction
Background: Free radicals and reactive oxygen species(ROS) are the most important factors in formation of oxidative stress reaction. Now, radical damage has been suggested to contribute to a wide variety of diseases such as Alzheimer, atherosclerosis and cancer. Transition metal ions in the presence of the various biomolecules produce these active compounds. The aim of this study is introducing...
متن کاملOn-demand cellular uptake of cysteine conjugated gadolinium based mesoporous silica nanoparticle with breast cancer-cells
Design, synthesis, and conjugation of mesoporous silica nanoparticles (MSNs) with biomolecules is a matter of growing interest to enhance selective uptake of contrast agents like gadolinium (Gd3+) by cancer cells. Here, by targeting xc-cystine/glutamate antiporter system in breast cancer cells, conjugation of MSN-Gd3+ with cysteine is used to enhance cancer cellular uptake of Gd3+. Reactions de...
متن کاملTHE DISTURBANCES IN IRON TRANSPORT AND STORES AND TOTAL FREE RADICAL TRAPPING ABILITY OF BLOOD PLASMA IN BABIES WITH MITOCHONDRIAL ENCEPHALO MYOPATHIES
Babies with mitochondrial encephalomyopathies had higher ferritin levels than controls. Although plasma iron levels were simi lar in both groups, babies with mitochondrial encephalomyopathies had lower transferrin levels. Thiobarbituric acid reactive substances in plasma of babies with mitochondrial encephalomyopathies were higher thun in controls suggesting increased lipid oxidation. We s...

The modified glassy carbon electrode (GCE) was prepared with 6-amino-4-(3,4-dihydroxyphenyl)-3-methyl-1,4-dihydropyrano[2,3-c]pyrazole-5-carbonitrile (pyrazole derivative (AMPC)) and functionalized multi-walled carbon nanotubes. In this research, electrocatalytic activity of nanocomposite (AMPC/MWCNTs) has been studied by cyclic voltammetry (CV), differential pulse voltammetry (DPV) and chronoa...
